The actor and human rights lawyer are pledging US$500,000 to the March event, which they will also attend.
"Amal and I are so inspired by the courage and eloquence of these young men and women from Stoneman Douglas High School," George Clooney said in a statement.
"Our family will be there on March 24 to stand side by side with this incredible generation of young people from all over the country, and in the name of our children Ella and Alexander, we're donating US$500,000 to help pay for this ground-breaking event.
"Our childrens' lives depend on it."
The calls for better protection from gun violence in the US come after its latest mass shooting, in which a man with white supremacist ties killed 17 people in a Florida school.
The March for Our Lives "will take to the streets of Washington, DC, to demand their lives and safety become a priority, and that we end gun violence and mass shootings in our schools today".
Speaking with CBS, one of the Florida students, Alex Wind, said the March 24 event is the first march, but it will not be the last.
Other celebrities showing support for the March include Justin Bieber, Amy Schumer, Barbara Streisand and Bette Midler.
