While it is not yet known what the white powder in the envelope was, the New York Police Department (NYPD) has now revealed it was "non-hazardous".
The trio were taken to hospital at 10am on Monday (local time) for evaluation. The couple have five children together, but it is not yet known if they were there at the time.
The Secret Service and NYPD are now investigating.
The letter was addressed to Mr Trump Jr and was opened by his wife at their shared apartment in Manhattan, New York.
