Rose took to Instagram to express her joy over the new role, in which her version of the DC character will be an out lesbian.
"The Bat is out of the bag and I am beyond thrilled and honoured," she wrote.
"I'm also an emotional wreck... because this is a childhood dream."
The Orange Is The New Black star also played lesbian character Stella Carlin in the hit Netflix series, and herself identifies as gender fluid.
"This is something I would have died to have seen on TV when I was a young member of the LGBT community who never felt represented on TV and felt alone and different."
As well as appearing in the Arrowverse series alongside superheroes Supergirl, The Flash and Green Arrow, Rose's Batwoman is set to star in her own, stand-alone show.
Writer Caroline Dries describes the show's Kate Kane/Batwoman character as "an out lesbian and highly trained street fighter" who must overcome her own demons in order to become Gotham City's savior.
Rose finished her emotional statement on Instagram with: "Thank you everyone. Thank you god".
