The 53-year-old told the Daily Star the book will be called In The Shadows Of The Duchess, and will cover everything "from lullabies to lies".
"The world does not know truth total truth...[I'm] holding nothing back!" she said.
Samantha, who has multiple sclerosis, has made many public jabs at the Duke and Duchess of Sussex; calling Harry "a wuss" and claiming her sister "mistreats" people.
Now, she's complaining that she's experiencing online harassment from the public, another topic she plans to cover in her book.
"On top of it I am dealing with cyber-stalking and cyber-bullying with little Twitter trolls," she said.
"They're kind of nasty, sending out banners and propaganda about me that's disparaging and not true."
Last month, Samantha was seen handing over a letter to a guard at Kensington Palace, before being turned away in her wheelchair.
She's previously made pleas for Meghan to reunite with their father, Thomas Markle, who claims he hasn't spoken to his daughter since he pulled out of the royal wedding at the last minute.
