The trio, who rose to fame after appearing in Disney Channel's Camp Rock, made the announcement with James Corden in a teaser for his 'Carpool Karaoke' feature.
One by one, Joe Jonas, Nick Jonas and Kevin Jonas all joined Corden in the car before confirming the reunion.
"Wait, hang on, is what I think is happening, is it happening?" the Late Late Show host asked.
"Yep, we're back!" the band replied.
"It's nice to be able to finally tell somebody," Nick said.
"We've kept this a secret now for almost seven, eight months."
The boy band also sang a snippet of their new single, 'Sucker', their first release together in five years.
"This is basically our first performance back," Kevin said of their 'Carpool Karaoke' appearance.
"We've got to dust off the cobwebs, here we go."
