Eight more people, which Newshub understands were on the bus, have minor injuries, St John said.
The road is closed at the intersection of Gowing Dr and Cruickshank Cres.
The serious crash unit is investigating.
Auckland Transport said it is aware of an incident involving a bus and car in Meadowbank earlier today.
"Our thoughts are with everyone affected, including AT customers on board, and we will be working closely with our bus operator and police as this issue is investigated," a spokesperson said.
Nigel Piper, Go Bus' chief operating officer, confirmed in a statement that one of its Auckland buses operating Route 782 collided with a car, leading to the death of one of the car occupants and serious injury to another.
He acknowledged that a number of passengers on the bus were injured before offering regards to them and the loved ones of all who have been affected by the accident.
"Our thoughts go out to the family of the deceased and those who were injured," he said.
"Go Bus will be supporting our driver, who is obviously quite shaken by the accident, and we will be working with police as they complete their investigation."
