Ko leads American Nelly Korda and Denmark's Koerstz Madsen by two shots, with a group of seven players a further shot back, tied for fourth.
The 23-year-old is on the hunt for her first tournament victory in three years, but showed glimpses of her best form in 2020, putting herself in contention on the final day several times, with nine top-20 finishes.
Ko didn't miss a cut during the 2020 season and starts this year ranked 29th in the world.
