The world No.3 has brought unconvincing form into the tournament at Baltusrol in Springfield, New Jersey and continued that slump with a three-over-par 74 over an opening round that featured five bogeys and two birdies.
Her score leaves her eight shots behind leader Lee-Anne Pace of South Africa, who holds a one-stroke advantage over Canadian Brooke Henderson, and Chinese pair Xiyu Lin and Ruoning Yin.
Tied at 61st, Ko sits inside the project cutline of top-65 players, plus ties.
